Descripción
Loxo@Lilly Oncology and CNIO are collaborating on the discovery of new targets for the development of potential cancer therapeutics. We are seeking to recruit a Laboratory Research Associate. The candidate will integrate in a group of 10 scientists at CNIO to evaluate and validate new cancer targets in the emerging areas of Chromosomal Inestability, DNA Repair, Tumor Metabolism, Immunotherapy, Cell signaling and Translational research.
Selected candidates will work in collaboration with several investigators for the design and execution of experiments towards target validation in the following research disciplines:
• Molecular biology: experiments involving molecular biology particularly CRISPR technology including CRISPR library screening, cloning in lentiviral and retroviral expression systems, gene silencing using shRNAs and the CRISPR system, and protein analysis mostly Western blot and IP techniques.
• Cellular biology: Tumor cell culture and primary culture including the preparation of mouse embryonic fibroblast, luciferase reporter assays, confocal immunofluorescence, high-throughput screening.
• In vivo work: candidates will need to demonstrate experience in animal husbandry, mouse models, with special emphasis in GEM models, xenografts/allografts models both classical and orthotopic as well as in vivo imaging.
• Bioinformatics and biostatistics: Knowledge in the use of public data bases, data mining and analysis.
• Immunohistochemistry: sample handling and analysis and imaging analysis.
• Bioenergetics: Seahorse analysis, metabolic flows by LC-MS and RMN.

Criterios de evaluación
At least 2 years of practical experience in the following areas:
• Molecular biology: gene silencing, library screening, PCR, isolation of nucleic acids and cloning, Western blot, immunoprecipitation and histology tools to analyze tissues from xenograft mouse models.
• Experience in cell culture techniques: Cell culture of tumor cells lines as well as primary culture of mouse embryonic fibroblast, human PBMC and mouse immune cells, ELISA, Immunofluorescence, Flow cytometry techniques and immunophenotyping.
• Husbandry of mouse colonies and animal handling particularly administration of compounds using different protocols and sampling of biological fluids, animal necropsy, in vivo imaging. It is mandatory to have finished the course in animal experimentation levels A, B & C.
• Immunohistochemistry and image analysis.
• Bioenergetics: Seahorse analysis, metabolic flows by LC-MS and RMN.
• Fluent in English.
